Zusammenfassung: | <jats:p><jats:bold>The Zip‐reaction: A New Method for the Synthesis of Macrocyclic Polyaminolactams</jats:bold></jats:p><jats:p>The 21‐ and 25‐membered aminolactams <jats:bold>11</jats:bold> and <jats:bold>25</jats:bold> were synthesized from the 13‐membered lactam <jats:bold>4</jats:bold>. To introduce the ring enlargement unit (a propylamino group) <jats:bold>4</jats:bold> was <jats:italic>N</jats:italic>‐alkylated using acrylonitrile and the resulting product hydrogenated. Repetition of this reaction sequence gave <jats:bold>3</jats:bold>, which was converted in the presence of base in 90% yield to the ring‐enlarged macrocyclic base <jats:bold>11</jats:bold> (<jats:italic>Scheme 2</jats:italic>). In a similar but stepwise synthesis consisting of two separate ring‐enlargement reactions <jats:bold>4</jats:bold> was transformed to <jats:bold>11</jats:bold> <jats:italic>via</jats:italic> <jats:bold>13</jats:bold> (<jats:italic>Scheme 4</jats:italic>). Introducing three ringenlargement units into <jats:bold>4</jats:bold> the 25‐membered aminolactam <jats:bold>25</jats:bold> was synthesized in 84% yield (<jats:italic>Scheme 5</jats:italic>). The mechanism of the ring‐enlargement reaction is given in <jats:italic>Scheme 3</jats:italic>. In comparison to a zip‐fastener or zipper this reaction is called “zipreaction”.</jats:p> |
